In order to realize the controllable electroporation of cell membrane under pulsed electric field
propidium iodide(PI) and Sybr GreenⅠwere used to perform fluorescent staining experiment of Chlorella vulgaris based on THU-PEF4(home-made high-voltage pulse power supply) system. The effects of equivalent pulse energy density and electric field intensity on the perforation rate of Chlorella cells were qualitatively studied under a laser confocal microscope. Besides
counting analysis of punctured Chlorella vulgaris cells combining with the Flow Cytometry was carried out. The effects of equivalent pulse energy density and electric field intensity on the perforation rate of Chlorella cells were quantitatively studied. Experimental r
esults show that the electroporation rate of Chlorella vulgaris increases with the increase of electric field strength under the laser confocal microscope. The proportion of irreversible electroporation of the Chlorella cells increases by about 15% when the electric field strength improves by 0.4
0
.5 MV/m under the equivalent pulse energy density of around 230 k J/L in the range of 2.5
4
MV/m. Chlorella cells perforation rate exceeds 89% when the equivalent pulse energy density reaches 230 k J/L under the electric field strength of 4 MV/m. The perforation ratio of Chlorella cells is linear with the electric field intensity within a certain range
showing the tendency to moderate after the first increase with the increase of the equivalent impulse energy density. Therefore
the controllable electroporation of Chlorella vulgaris cell membrane under pulsed electric field can be realized by controlling the electric field intensity and equivalent pulse injection energy density.
