In order to investigate the killing effect of low-intensity nanosecond pulsed electric field combined with multi-walled carbon nanotubes on cells
the CCK-8 detection method and Annexin V/PI double dyeing method based on the toxicity test of multi-walled carbon nanotubes were introduced to study the effect of different amplitude nanosecond pulsed electric fields on the activity and apoptosis necrosis rate of A375 cells after introducing multi-walled carbon nanotubes. Studies have shown that when the mass concentration of multi-walled carbon nanotubes is less than or equal to 200 mg/L
the multi-walled carbon nanotubes have no toxic effects on cells; meanwhile
only when the mass concentration exceeds 300 mg/L
the induced cytotoxicity is dose-dependent; in other words
the higher the mass concentration is
the stronger the toxicity to cells will be. When the fixed frequency is 1 Hz
the pulse width is 300 ns
and the number of pulses is 100
the cell mortality rate
apoptosis rate
and necrosis rate will increase with the increase of field strength. The introduction of multi-walled carbon nanotubes does not affect the above-mentioned variation law
but significantly improves the corresponding killing effect
and can improve the electrical safety in clinical treatment while maintaining the advantages of nanosecond pulse electric field treatment of tumor.
