Abstract
The synergistic manner of high-pressure narrow pulse and low-pressure wide pulse can significantly enhance the killing effect of irreversible electroporation(IRE) and expand the ablation volume. In order to deeply study the killing mechanism of synergistic pulse-enhanced irreversible electroporation
we designed an all-solid-state pulse generator topology that can flexibly generate high-voltage nanosecond and low-voltage microsecond cooperative pulse sequence. Based on field programmable gate array
a solid-state switch control platform was built to meet the requirements of controlling the timing requirements; finally
a set of all solid-state pulse generators with flexible generation of coordinated pulses were developed
and the performance tests and experimental verification of the prototype were carried out. The results show that the pulse generator can effectively generate synergistic pulse sequences and enhance cell killing effect. The nanosecond pulse parameters that can be output by the pulse source are as follows: voltage amplitude 0~10 kV adjustable
pulse width 100 ns~1 μs adjustable
and current amplitude up to 100 A under 100 Ω load; meanwhile
the microsecond pulse parameters are as follows: voltage amplitude 0~3 kV adjustable
pulse width 5~100 μs adjustable
and current amplitude up to 30 A under 100 Ω load
and nanosecond and microsecond pulse interval can be adjusted arbitrarily. The experimental results show that the output parameters and prototype performance of the all-solid-state synergistic pulse generator meet the requirements of collaborative pulse-induced IRE-related biological experiments
which lays a hardware foundation for further study of the bioelectrical effects of synergistic pulses.
